A modified antibody in which a plurality of Fc domains are tandemly linked to the heavy chain C terminus and a modified antibody in which Fc domains are tandemly linked through a spacer are produced, and the affinity, CDC activity, ADCC activity against an Fc receptor were measured. Although it has been reported that even if a plurality of Fc domains were linked, the CDC activity was not enhanced, the modified antibody of the invention exhibited an enhanced ADCC activity. According to the method of the present invention, it becomes possible to provide an antibody preparation having a high therapeutic effect.
L'invention a pour objet un anticorps modifié dans lequel une pluralité de domaines Fc sont liés en tandems à la terminaison C de la chaîne lourde et un anticorps modifié dans lequel des domaines Fc sont liés en tandems par un espaceur, tandis que l'affinité, l'activité CDC et l'activité ADCC envers un récepteur Fc sont mesurées. Bien qu'aucune augmentation de l'activité CDC n'ait été rapportée, même lorsqu'une pluralité de domaines Fc sont liés, l'anticorps modifié objet de l'invention a fait état d'une activité ADCC accrue. Le procédé objet de l'invention permet d'obtenir une préparation d'anticorps à fort effet thérapeutique.
