The immunogenicity of the influenza virus hemagglutinin (HA) molecule may be increased by substitutions of amino acids in the HA sequence. The substitution of specific HA residues, such as asparagine at position 223 of H5 HA, increase the sensitivity of the hemagglutinin inhibition (HI) assay by altering receptor specificity and/or antibody-antigen binding. HA molecules containing such substitutions will be useful in the development of diagnostic reference viruses and improved influenza vaccines.
Selon cette invention, l'immunogénicité de la molécule d'hémagglutinine (HA) du virus de la grippe peut être accrue par des substitutions d'acides aminés dans la séquence HA. La substitution de résidus HA spécifiques, tels que l'asparagine en position 223 de l'hémagglutinine H5, accroît la sensibilité du dosage d'inhibition de l'hémagglutinine (HI) du fait qu'elle modifie la spécificité du récepteur et/ou la liaison anticorps-antigène. Les molécules HA contenant de telles substitutions peuvent être utilisées pour développer des virus de référence de diagnostic et améliorer des vaccins contre la grippe.
