Compositions and methods are provided for the clinical assessment, diagnosis, and treatment of multiple sclerosis. The compositions of the invention are molecules related to the human proteolipid protein (PLP) and/or human myelin basic protein (MBP), and include nucleic acids and polypeptides. The nucleic acid molecules of the invention are useful in the production of modified PLP polypeptides and modified MBP polypeptides, such polypeptides being useful for assaying T cells for responsiveness to PLP and MBP epitopes. The polypeptides of the invention are also useful as therapeutic agents that act by inducing T cell responses, including anergy and apoptosis, as a means of treating multiple sclerosis.
L'invention porte sur des compositions et des procédés permettant l'évaluation clinique, le diagnostic et le traitement de la sclérose en plaque. Les compositions selon l'invention, qui consistent en des molécules liées à la protéine protéolipidique (PLP) et/ou la protéine myélinique de base (MBP) humaines, comportent également des acides nucléiques et des polypeptides. Les molécules d'acide nucléique de l'invention ont montré leur utilité dans la production de polypeptides PLP et MBP modifiés, ceux ci s'avérant utiles pour éprouver la sensibilité de lymphocytes T à des épitopes de la PLP et de la MBP. Les polypeptides selon cette invention sont également utiles comme agents thérapeutiques qui agissent en induisant des réponses de lymphocytes T, anergie et apoptose comprises, comme moyen de traitement de la sclérose en plaque.
