The invention herein includes a general method for reducing or eliminating a toxic effect of transgenic defensin expression in a host plant. The invention also includes a method of modifying a nucleic acid encoding a defensin, a nucleic acid modified thereby and a modified defensin encoded by the modified nucleic acid sequence. The invention also includes a transgenic plant containing and expressing the modified defensin-coding nucleic acid sequence, the plant exhibiting reduced or eliminated toxic effects of defensin, compared with otherwise comparable transgenic plants expressing an unmodified defensin. The modified defensin is termed a chimeric defensin having a mature defensin domain of a first plant defensin combined with a C-terminal propeptide domain of a second plant defensin or a vacuolar translocation peptide.
La présente invention concerne un procédé général pour réduire ou éliminer un effet toxique d'expression de la défensine transgénique dans une plante hôte. L'invention concerne également un procédé de modification d'acide nucléique codant pour une défensine, un acide nucléique modifié de cette façon et une défensine modifiée codée par une séquence d'acides nucléiques modifiée. L'invention concerne en outre une plante transgénique contenant et exprimant la séquence d'acides nucléiques codant pour la défensine modifiée, la plante présentant des effets toxiques de défensine réduits ou nuls par rapport à des plantes transgéniques autrement comparables exprimant une défensine non modifiée. La défensine modifiée est appelée défensine chimérique et est dotée d'un domaine de défensine mature d'une première défensine de plante combiné à un domaine propeptidique C-terminal d'une seconde défensine de plante ou d'un peptide de translocation vacuolaire.
