A modified polysaccharide, in particular a modified Neisseria meningitides serogroup A polysaccharide, which retains immunogenicity but has improved stability. Typically, the modified polysaccharide is prepared by reacting a capsular polysaccharide, or oligosaccharide fragment thereof, with a bifunctional reagent such as CDI, followed by reaction with an amino compound, such dimethylamine. Modified polysaccharide-protein conjugates and vaccines prepared from such conjugates are also described.
L'invention concerne un polysaccharide modifié, et notamment un polysaccharide modifié de Neisseria meningitidis du sérogroupe A, permettant de conserver l'immunogénicité tout en présentant une stabilité améliorée. Habituellement, ce polysaccharide modifié est préparé par réaction d'un polysaccharide capsulaire, ou d'un fragment oligosaccharidique de ce dernier, avec un réactif bifonctionnel tel que CDI, puis par réaction avec un composé amino tel que la diméthylamine. L'invention concerne également des conjugués polysaccharide-protéine et des vaccins préparés à partir de ces conjugués.
