The invention concerns novel yeast strains, methods and genetic constructs for preparing them, and their use for synthesising or modifying steroid compounds. More particularly, the invention concerns strains having reduced 20.alpha.-HSD activity, in particular by modification of the GCY1 and/or YPR1 genes. The inventive yeast strains enable to improve the efficacy of synthesis or to increase selectivity or yields of the process, as well as the quality of the final product. The inventive strains, methods and compounds are useful for researching, developing and producing products with therapeutic or prophylactic activity, in a human or an animal, in particular steroid derivatives.

La présente invention concerne de nouvelles souches de levure, des procédés et constructions génétiques pour leur préparation, ainsi que leur utilisation pour la synthèse ou la modification de composés stéroïdiens. Plus particulièrement, l'invention décrit des souches présentant une activité de type 20.alpha.-HSD diminuée, notamment par modification des gènes GCY1 et/ou YPR1. Les souches de levure de l'invention permettent d'améliorer l'efficacité de la synthèse ou d'augmenter la sélectivité ou les rendements du procédé, ainsi que la qualité du produit final. Les souches, procédés et composés de l'invention sont utiles dans la recherche, le développement et la production de produits à activité thérapeutique ou prophylactique, chez l'homme ou l'animal, notamment de dérivés stéroïdiens.
