It is known that proteins, or parts of them, may be rendered non- or less immunogenic to humans or other species by identifying one or more potential T cell epitopes and eliminating them by amino acid modification. Conventionally, certain epitopes may be retained in a protein sequence if the peptides constituting such epitopes are present in endogenous human protein, since they would be recognised as "self". However, it has now been found that even self epitopes may give rise to immune reactions. The invention provides for their elimination, for example by recombinant DNA technology, to render them more useful for administration to humans, for example for therapeutic or diagnostic purposes.

On sait qu'il est possible de rendre certaines protéines ou de leurs parties non, ou moins, immunogènes pour l'homme ou d'autres espèces, en identifiant un ou plusieurs épitopes potentiels de cellules T et en les éliminant par modification d'acides aminés. Il est admis que certains épitopes puissent être retenus dans une séquence de protéine si les peptides les constituant sont présents dans une protéine humaine endogène; on les qualifie alors d'"auto-épitopes". On a néanmoins trouvé que même les auto-épitopes peuvent donner lieu à des réactions immunitaires. L'invention porte sur leur élimination, par exemple au moyen d'une technique de recombinaison d'ADN qui les rend plus utilisables en vue de leur administration à l'homme, notamment à des fins thérapeutiques ou diagnostiques.
