The present invention provides improved N-hybrid assays. In particular, the present invention provides an improved reverse N-hybrid assays comprising modulating the amount of a substrate of a reporter gene and/or the amount of a reporter gene thereby enhancing cell death in the absence of a peptide inhibitor of a DNA- protein or protein- protein interaction and/or enhancing cell survival in the presence of a peptide inhibitor of a DNA-protein or protein interaction. Furthermore, the present invention provides an improved forward N-hybrid assay comprising modulating the amount of a reporter gene thereby enhancing cell death in the absence of a heterologous peptide or protein capable of binding to the DNA or protein in a cell and enhancing cell survival in the presence of a heterologous peptide or protein capable of binding to the DNA or protein in a cell.

L'invention concerne des essais améliorés à n hybrides. Notamment, l'invention concerne un essai inverse à n hybrides amélioré qui comprend la modulation de la quantité d'un substrat d'un gène rapporteur et/ou de la quantité d'un gène rapporteur, ce qui permet d'accroître la mort cellulaire en l'absence d'un peptide inhibiteur d'une interaction ADN-protéine ou protéine-protéine, et/ou d'accroître la survie cellulaire en présence d'un peptide inhibiteur d'une interaction ADN-protéine ou protéine-protéine. En outre, l'invention concerne un essai direct à n hybrides amélioré qui comprend la modulation de la quantité d'un gène rapporteur, ce qui permet d'accroître la mort cellulaire en l'absence d'un peptide ou d'une protéine hétérologue susceptible de se lier à l'ADN ou à la protéine dans une cellule et permet d'accroître la survie cellulaire en présence d'un peptide ou d'une protéine hétérologue susceptible de se lier à l'ADN ou à la protéine dans une cellule.
