The present invention relates to novel molecules (Fc variants) comprising at least one antigen binding region and an Fc region that further comprises a modified hinge which alters the binding of Fc to one or more Fc ligand (e.g., Fc.gamma.Rs) and/or modulates effector function. More specifically, this invention provides Fc variants that have modified binding affinity to one or more Fc.gamma.R and/or CIq. Additionally, the Fc variants have altered antibody-dependent cell-mediated cytotoxicity (ADCC) and/or complement dependent cytotoxicity (CDC) activity. The invention further provides methods and protocols for the application of said Fc variants particularly for therapeutic purposes.
La présente invention concerne de nouvelles molécules (variants Fc) comprenant au moins une région de liaison à un antigène et une région Fc comprenant une charnière modifiée qui altère la liaison Fc avec au moins ligand Fc (par ex., Fc?Rs) et/ou module la fonction d'effecteur. Plus spécifiquement, cette invention a trait à des variants Fc qui ont modifié l'affinité de liaison vis-à-vis de Fc?R et/ou CIq. En outre, les variants Fc présentent une cytotoxicité altérée à médiation cellulaire dépendante des anticorps et/ou une activité de cytotoxicité altérée dépendante de complément. Cette invention a aussi trait à des méthodes et des protocoles destinés à l'application desdits variants Fc, notamment, à des fins thérapeutiques.
