This invention relates to modulating Ror activity (e.g., Ror2 protein activity) and/or 14-3-3 .beta. to affect bone formation or resorption. The invention further relates to compositions and methods for the screening, diagnosis and development of therapies for bone-related disorders such as osteoporosis and bone fracture. Antibodies and antibody fragments directed to Ror2 protein are particularly useful in causing dimerization of Ror2 proteins, thereby leading to the activation of Ror2.
