The present invention relates to a chemokine construct comprising a scFV anti- CD4 and a RANTES chemokine or a functional fragment thereof, whereby said chemokine construct has the structural domain form of (a) RANTES-linker-(VH) anti-CD4-linker-(VL) anti-CD4; or (b) (VL) anti-CD4-linker-(VH) anti-CD4- linker-RANTES, or (c) (VH) anti-CD4-linker-(VL) anti-CD4-linker-RANTES, whereby in the alternatives (b) and (c), the linker between (VH) anti-CD4 and RANTES or between (VL) anti-CD4 and RANTES does not terminally comprise a methionine, a lysine or an isoleucine directly adjacent to the first amino acid of RANTES. Furthermore, polynucleotides encoding these chemokine constructs as well as vectors comprising these polynucleotides are disclosed. The invention also relates to specific uses of these constructs and to pharmaceutical compositions.

L'invention porte sur un gène hybride de chémokine comprenant un scFV anti-CD4 et une chémokine RANTES ou l'un de ses fragments fonctionnels, ledit gène hybride de chémokine présentant la forme de domaine structurel suivante; (a) RANTES-lieur-(VH) anti-CD4-lieur-(VL) anti-CD4; ou (b) (VL) anti-CD4-lieur-(VH) anti-CD4-lieur-RANTES, ou (c) (VH) anti-CD4-lieur-(VL) anti-CD4-lieur-RANTES, dans les variantes (b) et (c), le lieur compris entre (VH) anti-CD4 et RANTES ou entre (VL) anti-CD4 et RANTES ne comporte pas en position terminale de méthionine, de lysine ou de isoleucine jouxtant directement le premier acide aminé de RANTES. L'invention porte de plus sur des polynucléotides codant pour ces gènes hybrides de chémokine ainsi que sur des vecteurs comprenant lesdits polynucléotides; elle porte en outre sur des utilisations spécifiques de ces gènes hybrides et sur des préparations pharmaceutiques.
