An electrophoresis apparatus is generally disclosed for sequentially analyzing a single sample or multiple samples having one or more analytes in high or low concentrations. The apparatus (10) comprises a relatively large-bore transport passage (16) which intersects with a plurality of small-bore separation passages (28, 30, 32) and includes a valve system (18). Analyte concentrators (34, 36, 38), having antibody-specific (or related affinity) chemistries, are stationed at the respective intersections of the transport passage (16) and separation passages (28, 30, 32) to bind one or more analytes of interest. The apparatus allows the performance of two or more dimensions for the optimal separation of analytes. The apparatus may also include a plurality of valves surrounding each of the analyte concentrators to localize each of the concentrators to improve the binding of one or more analytes of interest.
De manière générale, la présente invention a trait à un appareil d'électrophorèse pour l'analyse séquentielle d'un échantillon unique ou d'une pluralité d'échantillons comportant un ou des analytes en des concentrations élevées ou faibles. L'appareil comporte un passage de transport à alésage relativement large d'intersection avec une pluralité de passages de séparation à alésage de taille réduite et comprend un système de soupapes. Des concentrateurs d'analytes, comprenant des solutions chimiques spécifiques d'anticorps (ou d'affinité liée), sont disposés aux intersections respectives pour la liaison d'un ou des analytes d'intérêt.
