ABSTRACT A particle analyzing method and apparatus for white blood cell (leukocyte) differentiation by optical and electronic measurements, without the use of dyes, on a flowing stream of leukocytes. A hydrodynamically focused stream of the leukocytes, in native condition, is passed into and through a beam of radiant energy in a flow cell, whereby the beam is uniquely scattered by different ones of the subpopulations of the leukocytes. The scattered light impinges upon light collection means disposed adjacent the flow cell. For eosinophil detection, the scattered light is collected at angular positions from about 10° to 70° relative to the axis of the beam. Other of the leukocyte subpopulations are differentiated relative to the eosinophils and each other by optical and/or electronic measurements, especially including the Coulter Principle of particle analysis.
