The development of miniaturized chromatographic systems localized within individual polymer microspheres and their incorporation into a bead-based cross-reactive sensor array platform is described herein. The integrated chromatographic and detection concept is based on the creation of distinct functional layers within the microspheres. In this first example of the new methodology, complexing ligands have been selectively immobilized to create "separation" layers harboring an affinity for various analytes. Information concerning the identities and concentrations of analytes may be drawn from the temporal properties of the beads' optical responses. Varying the nature of the ligand in the separation shell yields a collection of cross-reactive sensing elements well suited for use in array-based micro-total-analysis systems.
L'invention concerne le développement de systèmes chromatographiques miniaturisés localisés dans des microsphères polymères individuelles et leur incorporation dans une plate-forme de mosaïques de capteurs à réactivité croisée à base de billes. Le concept de chromatographie et de détection intégrées est basé sur la création de couches fonctionnelles distinctes dans les microsphères. Dans le premier exemple de cette nouvelle méthodologie, des ligands de complexage ont été sélectivement immobilisés afin de créer des couches de séparation hébergeant une affinité pour des analytes variés. Des informations concernant les identités et les concentrations des analytes peuvent être tirées des propriétés temporelles des réponses optiques des billes. La nature du ligand variant dans la coque de séparation produit une collection d'éléments de détection à réactivité croisée appropriés pour être utilisés dans des systèmes de microanalyse globale.
