Compositions for coating biological and non-biological surfaces, which minimize or prevent cell-cell contact and tissue adhesion, and methods of preparation and use thereof, are disclosed. Embodiments include polyethylene glycol/polylysine (PEG/PLL) block or comb-type copolymers with high molecular weight PLL (greater than 1000, more preferably greater than 100,000); PEG/PLL copolymers in which the PLL is a dendrimer which is attached to one end of the PEG; and multilayer compositions including alternating layers of polycationic and polyanionic materials. The multi-layer polymeric material is formed by the ionic interactions of a polycation and a polyanion. The molecular weights of the individual materials are selected such that the PEG portion of the copolymer inhibits cellular interactions, and the PLL portion adheres well to tissues. The compositions and methods are useful, for example, in inhibiting formation of post-surgical adhesions, protecting damaged blood vessels from thrombosis and restenosis, and decreasing the extent of metastasis of attachment-dependent tumor cells. The compositions and methods are also useful for coating non-biological surfaces such as metallic surfaces.

On décrit des compositions de recouvrement de surfaces biologiques ou non biologiques, qu'on utilise pour réduire au minimum ou empêcher un contact entre cellules ou une adhésion tissulaire, et des méthodes de préparation et d'utilisation desdites compositions. Dans une forme de réalisation, on décrit un bloc d polyéthylèneglycol/polylysine (PEG/PLL) ou des copolymères en peigne présentant un PLL de masse moléculaire élevée (supérieure à 1000, ou de préférence supérieure à 100.000); des copolymère PEG/PLL dans lesquels la PLL est un dendrimère fixé à une extrémité du PEG; et des compositions en couches multiples incluant des couches alternées de matériaux poylcationiques et polyanioniques. Le matériau polymérique à couches multiples est formé par les interaction ioniques d'un polycation et d'un polyanion. Les masses moléculaires des matériaux pris séparément sont choisies de telle sorte que la partie PEG du copolymère inhibe les interactions cellulaires et la partie PLL présente une bonne adhérence aux tissus. Lesdites compositions et méthodes son efficaces, par exemple, pour inhiber l'apparition d'adhésions post-chirurgicales, pour protéger des vaisseaux sanguins endommagés contre la thrombose ou la resténose, et pour ralentir l'évolution de la métastase de cellules cancéreuses dépendantes de la fixation.
