A soluble multimeric polypeptide or protein is disclosed that is able to inhibit interaction of leukocyte Fcy receptors (Fc.gamma.R) and immunoglobulin G (IgG). The protein or polypeptide comprises two or more Fc binding regions linked in a head to tail arrangement, at least one of which is derived from an Fc.gamma.R type receptor, and an Fc domain of an immunoglobulin which has been modified to reduce or prevent binding to the said Fc binding regions and/ or to alter effector function. Also described are polynucleotide molecules encoding the polypeptide or protein and the use thereof in methods of treating a subject for an immune- complex (IC)-mediated inflammatory disease.
Cette invention concerne un polypeptide ou une protéine multimère soluble qui est capable d'inhiber l'interaction des récepteurs Fc.gamma. des leucocytes (FC.gamma.R) et des immunoglobulines G (IgG). La protéine ou le polypeptide comprend deux régions de liaison du Fc ou plus, liées selon une disposition tête à queue, au moins l'une d'entre elles étant dérivée d'un récepteur de type Fc.gamma.R, et un domaine Fc d'une immunoglobuline qui a été modifié pour réduire ou empêcher la liaison aux dites régions de liaison du Fc et/ou pour modifier la fonction effectrice. Cette invention concerne également des molécules de polynucléotides codant pour le polypeptide ou la protéine et leur utilisation dans des procédés de traitement d'un sujet pour une maladie inflammatoire médiée par des complexes immuns (CI).
