Multiple branch peptide constructions formed from peptides derived from the envelope transmembrane glycoprotein gp41 of HIV, and including from 2 to 16 branches of the peptide RQGYS or from 8 to 16 branches of the peptide RQGY, show increased receptor affinity and prevent cell-to-cell fusion. They have a direct virostatic effect. These MBPCs are able to neutralize in vitro the different steps of virus envelope/cell membrane fusion, and infected cell membrane/uninfected cell membrane fusion of several strains of HIV-1 and HIV- 2. These results open a potential use in treatment of HIV infection.
L'invention concerne des constructions peptidiques à ramifications multiples (MBCP) formées à partir de peptides dérivés de la glycoprotéine transmembranaire d'enveloppe gp41 du VIH, et qui comprennent entre 2 et 16 ramifications du peptide RQGYS ou entre 8 et 16 ramifications du peptide RQGY. Ces constructions présentent une affinité de récepteur améliorée, empêchent la fusion cellule à cellule, et ont un effet virostatique direct. Ces MBPC peuvent également neutraliser in vitro les différentes étapes de fusion d'enveloppe de virus/de membrane cellulaire, et de fusion de membrane cellulaire infectée/de membrane cellulaire non infectée de plusieurs brins de VIH-1 et de VIH-2. Ces résultats ouvrent une voie potentielle pour le traitement des infections par le VIH.
