Compositions and methods for making and using therapeutic formulations of multivalent hybrid polypeptides comprising immunogenic peptides of M proteins from various different serotypes of group A streptococci and antibodies thereto are provided. Also provided are nucleic acids encoding such hybrid polypeptides. The hybrid polypeptide formulations may be used, for example, in methods for treating or preventing a microbial infection and eliciting a protective immune response having broadly protective opsonic antibodies in the absence of tissue cross-reactive antibodies.
La présente invention concerne des compositions et des procédés de fabrication et d'utilisation de formulations thérapeutiques de polypeptides hybrides multivalents comprenant des peptides immunogéniques de protéines M en provenance de plusieurs sérotypes différents de streptocoques du groupe A, et des anticorps dirigés contre ces derniers. L'invention se rapporte également à des acides nucléiques codant ces polypeptides hybrides. Les polypeptides de l'invention peuvent être utilisés, par exemple, selon des procédés qui permettent de traiter ou prévenir une infection microbienne et de susciter une réponse immunitaire de protection par des anticorps opsoniques largement protecteurs en l'absence d'anticorps tissulaires de réaction croisée.
