A multiwell plate scanner comprises a detector for measuring an attribute of a sample which is scanned continuously over wells of a multiwell plate. A signal obtained during the scan may be sampled and digitized based on detector position over the multiwell plate. The scanner is also disclosed for scanning microarrays, bio-chips and areas of samples not having physical separations. The scanner may be used in a high throughput screening system comprising a storage and retrieval module, a sample distribution module, a reagent distribution module, and a detector which incorporates the scanner. The screening system may further comprise a transport module and a data processing and integration module for transporting samples between the components of the system and for controlling system operation. In a further aspect, the present invention comprises methods and apparatus to compensate for variations, inconsistencies and other differences in the dimensions, configuration and/or materials of the platform in which an assay is performed. Another aspect of the invention is a system and method for performing an assay to detect the effect of a reagent on a target. The present invention is also directed to compositions and therapeutics identified by the disclosed methods. A further aspect of the present invention is a method of testing a therapeutic for therapeutic activity and toxicology by identifying a compound using a method of the present invention and monitoring the toxicology and efficacy of the therapeutic in an in vivo model.
L'invention concerne un analyseur pourvu d'une plaque à cupules multiples. Cet analyseur comprend un détecteur destiné à mesurer un attribut d'un échantillon balayé en continu au-dessus des cupules d'une plaque à cupules multiples. Un signal obtenu pendant le balayage peut être échantillonné et numérisé sur la base de la position du détecteur au-dessus de la plaque à cupules multiple. L'analyseur permet également de balayer des jeux ordonnés de microéchantillons, des biopuces et des zones d'échantillons ne présentant pas de séparations physiques. Ledit analyseur peut être utilisé dans un système de criblage à débit élevé comprenant un module de stockage et de récupération, un module de distribution d'échantillons, un module de distribution de réactifs et un détecteur comprenant ledit analyseur. Ce système de criblage peut également comprendre un module de transport et un module d'intégration et de traitement de données servant à transporter des échantillons entre les composants du système et à commander le fonctionnement dudit système. Dans un autre mode de réalisation, la présente invention comprend des procédés et un dispositif destinés à compenser les variations, les irrégularités et d'autres différences de dimensions, de configuration et/ou de matériaux de la plateforme sur laquelle on procède à une analyse. Selon un autre aspect de l'invention, un système et un procédé permettent d'effectuer une analyse consistant à détecter l'effet d'un réactif sur une cible. La présente invention concerne également des compositions et des agents thérapeutiques identifiés au moyen des procédés susmentionnés. Cette invention se rapporte en outre à un procédé permettant de tester un agent thérapeutique de façon à en déterminer l'activité thérapeutique et la toxicité par identification d'un composé selon un procédé de l'invention et par contrôle de la toxicité et de l'efficacité dudit agent thérapeutique chez un spécimen in vivo.
