A novel mutant endonuclease V not exhibiting a nonspecific nucleic acid cleavage activity but showing a specific activity. The present invention relates to a mutant endonuclease V not exhibiting a nonspecific nucleic acid cleavage activity but having a specific activity, to a gene coding for the endonuclease V, to a recombinant DNA containing the gene, to a transformant or transductant containing the recombinant DNA, and to a method for producing the endonuclease V. The present invention also relates to a method of using the endonuclease V for nucleic acid cleavage, and to a reagent kit for detection or modification of a nucleic acid or gene that contains the endonuclease V.
L'invention porte sur une nouvelle endonucléase V mutante qui ne présente pas une activité de clivage non spécifique des acides nucléiques, mais dont l'activité est spécifique. La présente invention concerne une endonucléase V mutante qui ne présente pas d'activité de clivage non spécifique des acides nucléiques, mais une activité spécifique envers le gène codant l'endonucléase V, l'ADN recombinant contenant le gène, le transformant ou le transductant contenant l'ADN recombinant et une méthode pemettant de produire l'endonucléase V. Elle porte aussi sur une méthode faisant appel à l'endonucléase V pour cliver des acides nucléiques, et sur une trousse de réactifs destinée à la détection ou à la modification d'un acide nucléique ou d'un gène qui contient l'endonucléase V.
