The invention concerns a polypeptide having a uracil phosphoribosyl transferase (UPRTase) activity derived from a native UPRTase by mutation of one or several residues of said UPRTase. The invention also concerns a nucleotide sequence coding for said UPRTase mutant, a vector for expressing the latter, a viral particle and a host cell as well as a composition containing them. The invention further concerns their therapeutic use and a treatment method using them. The invention is particularly useful in the context of therapy by suicide genes in particular for treating proliferative and infectious diseases.

La présente invention a pour objet un polypeptide ayant une activité uracile phosphoribosyl transférase (UPRTase) dérivant d'une UPRTase native par mutation d'un ou plusieurs résidus de ladite UPRTase. Elle concerne également une séquence nucléotidique codant pour ce mutant UPRTase, un vecteur pour l'expression de cette dernière, une particule virale et une cellule hôte ainsi qu'une composition les comprenant. Enfin, elle vise également leur utilisation thérapeutique ainsi qu'une méthode de traitement les mettant en oeuvre. La présente invention est particulièrement utile dans le cadre de la thérapie par gènes suicides pour une application à l'égard notamment des maladies prolifératives et infectieuses.
