The invention relates to muteins of bilin-binding protein with a binding ability to digoxigenin and the fusion proteins of said muteins, a method for preparing said muteins and fusion proteins thereof and to their utilization for detecting or binding digoxigenin-labeled biomolecules. The invention especially relates to a polypeptide selected from the muteins of the bilin- binding protein, which is characterized in that (a) it can bind digoxigenin or digoxigenin conjugates; (b) it does not bind ouabain, testosterone and 4- aminofluorescein (c) at least one of the sequence positions 28, 31, 34, 35, 36, 37, 58, 60, 69, 88, 90, 95, 97, 114, 116, 125 and 127 of the bilin-binding protein has an aminoacid substitution. Due to their simple molecular structure, the inventive muteins provide advantages for production and utilization in comparison with antibodies against the digoxigenin group.
L'invention concerne des mutéines de la protéine liant la biline présentant une capacité de liaison avec la digoxigénine, ainsi que des protéines de fusion de telles mutéines. Elle concerne également des procédé de production de ces mutéines et leurs protéines de fusion, ainsi que leur utilisation pour déceler ou fixer des biomolécules marquées à la digoxigénine. L'invention concerne en particulier un polypeptide, sélectionné dans des mutéines de la protéine liant la biline. Ce polypeptide est caractérisé en ce que (a) il peut fixer la digoxigénine ou des conjugués de cette dernière, (b) il ne fixe pas la strophantine G, la testostérone et la 4-aminofluorescéïne, et (c) il présente, à au moins une des positions de séquence 28, 31, 34, 35, 36, 37, 58, 60, 69, 88, 90, 95, 97, 114, 116, 125 et 127 de la protéine liant la biline, une substitution d'acide aminé. En raison de leur structure moléculaire simple, les mutéines selon l'invention présentent des avantages sur le plan de la préparation et de l'utilisation, par rapport aux anticorps contre le groupe digoxigénine.
