Primers and probes can be used to detect nucleic acid from Mycobacterium in a sample and determine the species from which the nucleic acid originates. The primers amplify regions of the 16S ribosomal RNA gene and hybridize to regions conserved among species. Genus specific probes hybridize to sequences within the amplified region conserved among mycobacterial species, whereas the species speck probes hybridize to a variable region, so that the species identity can be uniquely determined. Consensus probes for detecting mycobacteria nucleic acids are provided which probes are not identical to any of the sequences of mycobacterial species.
