This invention is directed to substituted imidazole compounds which modulate the activity of KSP and are useful for the treatment of cancer and are represented by the formula (I) wherein: R1 is selected from the group consisting of aminoacyl, acylamino, carboxyl, carboxyl ester, aryl, and alkyl optionally substituted with hydroxy or halo; R2 is selected from the group consisting of hydrogen, alkyl, and aryl; R3 is selected from the group consisting of hydrogen and -X-A, wherein X is selected from the group consisting of-C(O)-, -C(S)-, -S(O)-, -S(O)2-, and -S(O)2-N(R)-, where R is hydrogen or alkyl; and A is selected from the group consisting of hydrogen, optionally substituted alkyl, optionally substituted alkoxy, optionally substituted aryl, carboxyl, carboxyl ester, aminoacyl, optionally substituted heteroaryl, optionally substituted heterocyclic, and optionally substituted cycloalkyl, wherein the optionally substituted groups are substituted with 1 to 4 substituents; R5 is selected from the group consisting of L-A1, wherein L is selected from the group consisting of-S(O)q- where q is one or two, and C1 to C5 alkylene optionally substituted with hydroxy, halo, or acylamino; and A1 is selected from the group consisting of aryl, substituted aryl, heteroaryl, substituted heteroaryl, heterocyclic, substituted heterocyclic, cycloalkyl, and substituted cycloalkyl; and one of either R6 or R7 is selected from the group consisting of heterocyclic, aryl and heteroaryl, all of which may be optionally substituted with -(R8)m where R8 is as defined herein and m is an integer from 1 to 3, and the other Of R6 or R7 is selected from the group consisting of hydrogen, halo, and alkyl; or R6 and R7 are both hydrogen; the other substituents are defined in the claims.
La présente invention a trait à des composés d'imidazole substitués modulateur de l'activité de la KSP et utiles pour le traitement du cancer, représentés par la formule (I), dans laquelle: R1 est choisi parmi le groupe constitué d'aminoacyle, d'acylamino, de carboxyle, d'ester de carboxyle, d'aryle, et d'alkyle éventuellement substitué par hydroxy ou halo; R2 est choisi parmi le groupe constitué d'hydrogène, d'alkyle et d'aryle; R3 est choisi parmi le groupe constitué d'hydrogène, et de -X-A, où X est choisi parmi le groupe constitué de -C(O)-, -C(S)-, -S(O)-, -S(O)2-, et -S(O)2-N(R)-, où R est hydrogène ou alkyle; et A est choisi parmi le groupe constitué d'hydrogène, alkyle éventuellement substitué, alcoxy éventuellement substitué, aryle éventuellement substitué, carboxyle, ester de carboxyle, aminoacyle, hétéroaryle éventuellement substitué, hétérocyclique éventuellement substitué, et cycloalkyle éventuellement substitué, où le groupes éventuellement substitués sont substitués avec 1 à 4 substituants; R5 est choisi parmi le groupe constitué de L-A1, où L est choisi parmi le groupe constitué de -S(O)q- où q est 1 ou 2, et d'alkylène en C1-C5 éventuellement substitué par hydroxy, halo, ou acylamino; et A1 est choisi parmi le groupe constitué d'aryle, d'aryle substitué, d'hétéroaryle, dhétéroaryle substitué, d'hétérocyclique, d'hétérocyclique substitué, de cycloalkyle, et de cylcoalkyle substitué; et un parmi R6 et R7 est choisi parmi le groupe constitué d'hétérocyclique, d'aryle et d'hétéroaryle, dont tous peuvent être éventuellement substitués par -(R8)m où R8 est tel que défini dans la description et m est un nombre entier de 1 à 3, et l'autre parmi R6 et R7 est choisi parmi le groupe constitué d'hydrogène, d'halo et d'alkyle; ou R6 et R7 sont tous deux hydrogène; les autre substituants sont tels que définis dans les revendications.
