A novel pyrimidine derivative, N-(2-amino-4,6- dichloropyrimidin-5-yl)formamide, which can be readily converted into nucleotide derivatives, is described, as is a process for its preparation. In the process for preparing N-(2-amino-4,6-dichloropyrimidin-5-yl)formamide of the formula: Image an aminomalonic ester of the general formula: Image or a salt thereof is cyclized, in a first step, with guanidine or a salt thereof in the presence of a base to give 2,5-diamino-4,6-dihydroxypyrimidine of the formula: Image or a salt thereof. The compound (III) then chlorinated, in a second step, with a chlorinating agent in the presence of an amide of the general formula: Image to give a 4,6-dichloropyrimidine of the general formula: Image The compound (V) is then reacted, in a third step, with an aqueous solution of a carboxylic acid of the general formula: Image to give the end product of formula I.
