An N-(3-pyrrolidinyl)benzamide derivative represented by general formula (I) or a pharmaceutically acceptable salt thereof and a medicine containing the same, having a potent and selective antagonism against dopamine D3 and/or D4 receptors and useful as a psychotropic agent, schizophrenia remedy, and so forth. In said formula, R1 represents hydrogen, lower alkyl, aralkyl, or three- to eight-membered cycloalkyl or cycloalkyl-lower alkyl; R2 represents four- to sixteen-membered bi- or tricyclic bridged hydrocarbon group; R3 represents lower alkoxy, amino, or mono- or di(lower alkyl)amino; R4 represents hydrogen, halogen, optionally hydroxylated lower alkyl, lower alkoxy, cyano, nitro, amino, mono- or di(lower alkyl)amino, acyl or -S(O)m-R5; R5 represents lower alkyl, amino or mono- or di(lower alkyl)amino; m represents 0, 1 or 2; X represents a bond, -O-, -S(O)n-, -NH- or -CONH-; and n represents 0, 1 or 2; provided when R1 represents cycloalkyl, the case where X represents -CONH-, R3 represents lower alkoxy and R4 represents halogen is excepted.

Cette invention se rapporte à un derivé de N-(3-pyrrolidinyl)benzamide, représenté par la formule générale (I), ou à un sel pharmaceutiquement acceptable de ce dérivé, ainsi qu'à un médicament contenant ce dérivé. Ce dérivé possède un pouvoir antagoniste puissant et sélectif contre les récepteurs de la dopamine D¿3? et/ou D¿4? et il peut servir comme agent psychotrope, comme remède contre la schizophrénie, etc. Dans la formule (I), R?1¿ représente hydrogène, alkyle inférieur, aralkyle, ou cycloalkyle ou cycloalkyle-alkyle inférieur ayant trois à huit éléments; R?2¿ représente un groupe hydrocarbure ponté bicyclique ou tricyclique ayant quatre à seize éléments; R?3¿ représente alcoxy inférieur, amino, ou amino mono(alkyle inférieur) ou di(alkyle inférieur); R?4¿ représente hydrogène, halogène, alkyle inférieur éventuellement hydroxylé, alcoxy inférieur, cyano, nitro, amino, amino mono(alkyle inférieur) ou di(alkyle inférieur), acyle ou -S(O)¿m?-R?5¿; R?5¿ représente alkyle inférieur, amino ou amino mono(alkyle inférieur) ou di(alkyle inférieur); m est égal à 0, à 1 ou à 2; X représente une liaison, -O-, -S(O)¿n?-, -NH- ou -CONH-; et n est égal à 0, à 1 ou à 2; à condition que, lorsque R?1¿ représente cycloalkyle, alors les cas où X représente -CONH-, R?3¿ représente alcoxy inférieur et R?4¿ représente halogène soient exclus.
