The invention provides exemplary devices and methods to facilitate the performance of assays. In one embodiment, one such device comprises a holding member (58) having a top surface, a bottom surface, and a plurality of holding locations (62) that are adapted to hold at least one article (66), such as a solid support and/or a cell. When within the holding locations, the articles (66) are preferably disposed below the top surface. A membrane (68) is positioned above the top surface of the holding member (58), and a pressure system is provided to apply positive pressure to the membrane (68) to force the membrane (68) against the top surface of the holding member (58). In this way, a seal may be provided between the holding locations (62).
