Nanoparticulate insulin compositions having effective an average particle size of less than about 5 microns are described. The compositions exhibit retention of insulin bioactivity, increased bioavailability, and increased consistency of bioavailability as compared to prior macro-sized insulin compositions. Methods of making nanoparticulate insulin compositions, dosage forms containing them, and the use of such formulations in insulin therapy are also described.

L'invention concerne des préparations d'insuline nanoparticulaires présentant une dimension moyenne des particules inférieure à 5 microns environ. Ces préparations permettent une rétention de la bioactivité de l'insuline, une augmentation de la biodisponibilité et une augmentation de l'uniformité de la biodisponibilité par rapport à des préparations d'insuline actuelles présentant des dimensions moyennes des particules supérieures. L'invention concerne également des méthodes de fabrication de préparations d'insuline nanoparticulaires, des formes pharmaceutiques renfermant ces préparations et l'utilisation de ces préparations pour l'insulinothérapie.
