A delivery device for and method of providing for delivery of substance to the central nervous system (CNS) of a subject, the delivery device comprising: a nosepiece unit (17) for insertion into a nasal airway (1) of a subject and comprising an outlet unit (21) which includes a nozzle (25) for delivering substance into the nasal airway of the subject; and a substance supply unit which is operable to deliver a dose of substance to the nozzle: wherein the delivery device is configured such that at least 30 % of the dose as initially deposited in the nasal airway is deposited in an upper posterior region of the nasal airway, thereby providing a CNS concentration of the substance, and hence CNS effect, which is significantly greater than that which would be predicted from a counterpart blood plasma concentration of the substance.
L~invention concerne un dispositif d~administration et une méthode pour permettre l~administration d~une substance au système nerveux central (SNC) d'un sujet, l~appareil de livraison comprenant : une unité nasale (17) à insérer dans une voie respiratoire nasale (1) d'un sujet et comprenant une unité de sortie (21) laquelle comprend une buse (25) pour délivrer une substance dans la voie respiratoire nasale du sujet ; et une unité d~alimentation en substance qui permet de délivrer une dose de substance à la buse. Ledit un dispositif d~administration est configuré de façon à ce qu~au moins 30% de la dose initialement déposée dans la voie respiratoire nasale soient déposés dans une région postérieure supérieure de la voie respiratoire nasale, arrivant ainsi à une concentration en substance dans le SNC, et donc à un effet sur le SNC, qui sont considérablement plus élevés que ceux attendu d'une concentration plasmatique sanguine équivalente de la substance.
