The present invention relates to an immunogenic composition or vaccine, Neisserial strain and process for making an oligosaccharide structure from these Neisserial strains. The immunogenic composition comprises an oligosaccharide structure prepared from a recombinant Neisserial strain, said Neisserial strain being incapable of expressing a functionally active form of a glycosyltransferase selected from a group consisting of: lipooligosaccharide glycosyl transferase A (LgtA), the Neisseria gonorrhoeae strain F62 amino acid sequence of which being SEQ ID NO: 3; lipooligosaccharide glycosyl transferase B (LgtB), the Neisseria gonorrhoeae strain F62 amino acid sequence of which being SEQ ID NO: 8; lipooligosaccharide glycosyl transferase C (LgtC), the Neisseria gonorrhoeae strain F62 amino acid sequence of which being SEQ ID NO: 4; lipooligosaccharide glycosyl transferase D (LgtD), the Neisseria gonorrhoeae strain F62 amino acid sequence of which being SEQ ID NO: 5; and lipooligosaccharide glycosyl transferase E (LgtE), the Neisseria gonorrhoeae strain F62 amino acid sequence of which being SEQ ID NO: 6, wherein the functionally active form of the glycosyltransferase catalyzes a reaction selected from the group consisting of: a) adding Gal .beta.1.fwdarw.4 to GlcNAc or Glc; b) adding GalNAc or GlcNAc .beta.1.fwdarw.3 to Gal; and c) adding Gal .alpha.1.fwdarw.4 to Gal.
La présente invention porte sur une préparation immunogène ou un vaccin, une souche de Neisseria et une méthode permettant d'obtenir un oligosaccharide à partir de ces souches de Neisseria. La préparation immunogène contient un oligosaccharide préparé à partir d'une souche de Neisseria recombinante, ladite souche de Neisseria étant incapable d'exprimer un forme active fonctionnelle d'une glycosyltransférase choisie parmi le groupe suivant : lipooligosaccharide glycosyl transférase A (LgtA), séquence d'acides aminés de la souche Neisseria gonorrhoeae F62 SEQ ID No 3; lipooligosaccharide glycosyl transférase B (LgtB), séquence d'acides aminés de la souche de Neisseria gonorrhoeae F62 SEQ ID No 8; lipooligosaccharide glycosyl transférase C (LgtC), séquence d'acides aminés de la souche de Neisseria gonorrhoeae F62 SEQ ID No 4; lipooligosaccharide glycosyl transférase D (LgtD), séquence d'acides aminés de la souche Neisseria gonorrhoeae F62 SEQ ID No 5; et lipooligosaccharide glycosyl transférase E (LgtE), séquence d'acides aminés Neisseria gonorrhoeae F62 SEQ ID No 6, où la forme fonctionnellement active de la glycosyltransférase catalyse une réaction choisie parmi les suivantes : a) ajouter Gal 1 4 à GlcNAc ou Glc; b) ajouter GalNAc ou GlcNAc 1 3 à Gal; et c) ajouter Gal 1 4 à Gal.
