A novel immunoglobulin-like glycoprotein is described which has been found to occur in high concentrations in the urine of patients with glomerulosclerosis of end-stage renal disease. This nephropathy-related protein is also found at a high frequency in the urine of patients with a high risk of developing kidney disease, as well as in the urine of kidney transplant recipients who are expriencing early signs of organ failure or rejection. Although this glycoprotein has several antigenic epitopes which are identical to those on normal human IgG molecules, two monoclonal antibodies have been developed which bind epitopes which are unique to this nephropathy-related immunoglobulin-like molecule. Immunometric assays have been developed which permit this nephropathy-related protein to serve as an early and specific marker for kidney-related diseases.
L'invention concerne une nouvelle glycoprotéine du type immunoglobuline dont la présence en fortes concentrations a été découverte dans l'urine de patients atteints de la glomérlulosclérose causée par la néphropathie au stade terminal. Cette protéine liée à la néphropathie se rencontre également à une fréquence élevée dans l'urine de patients présentant un risque élevé de maladie de reins, ainsi que dans l'urine de receveurs de greffe rénale connaissant des signes précoces de défaillance ou de rejet d'organe. Bien que cette glycoprotéine possède plusieurs épitopes antigéniques qui sont identiques à ceux présents sur les molécules d'IgG humaines normales, deux anticorps monoclonaux ont été mis au point et fixent des épitopes qui sont uniques à cette molécule de type immunoglobuline liée à la néphropathie.
