Disclosed is a new process for producing finasteride which involved treating the magnesium halide salt of 17.beta.-carboalkoxy-4- aza-5.alpha.-androst-1-en-3-one with t-butylamino magnesium halide, present in at least a 2:1 molar ratio to the ester, formed from t-butyl amine and an aliphatic/aryl magnesium halide at ambient temperature in an inert organic solvent under an inert atmosphere followed by heating and recovering the product finasteride. Also disclosed are two polymorphic crystalline Forms I and II of finasteride, and methods of their production. The process of the present invention is useful to obtain finasteride, a 5.alpha.-reductase inhibitor, in good yield. The polymorphic forms of finasteride are useful as 5.alpha.-reductase inhibitors, for treating hyperandrogenic disorders, including benign prostatic hyperplasia, acne, androgenic alopecia, including male pattern baldness, female hirsutism, prostatitis, and prostatic carcinoma.

Le présent document porte sur un nouveau procédé de production du finastéride qui consiste à traiter du sel de magnésium halogéné de 17.beta.-carboalkoxy-4-aza-5.alpha.-androst-1-ène-3- one avec le t-butylamine de magnésium halogéné, en présence d'ester dans une proportion molaire d'au moins 2:1, lequel est formé à partir du t-butylamine et d'un composé de magnésium aliphatique ou aryle à la température ambiante, dans un solvant organique inerte et une atmosphère inerte, puis par exposition à la chaleur avant la récupération du finastéride. On y divulgue de plus deux formes cristallines polymorphes (I et II) du finastéride, ainsi que leurs procédés de production. Le processus décrit dans la présente invention se révèle utile pour la fabrication du finastéride, un inhibiteur de 5.alpha-réductase, avec un rendement substantiel. Les formes polymorphes du finastéride sont utiles comme inhibiteurs de 5.alpha-réductase, pour le traitement de l'hyperandrogénie, de l'hypertrophie bénigne de la prostate, de l'acné, de la pelade, y compris la calvitie masculine, de l'hirsutisme féminin, de la prostatite et du carcinome prostatique.
