The present invention relates to new Bacillus sp. SY-414 that produced antifungal antibiotic, tri- hexocin, which was produced by the above Bacillus sp. SY-414 and its manufacturing method. New antifungal antibiotic represented by general formula (1), wherein each R denotes -H, -OH, -NH27 alkyl group of C1-C6, allyl group of C5- C20, or alkylene group of C2-C6, and subscript l, m, or n is an in- teger less than 3. The manufactur- ing method for the above antifungal antibiotic is as follows: new Bacil- lus sp. microorganism showing a high activity against plant fungi, especially Pyricularia oryzae and Pellicularia filamentosa, was iso- lated from soil and identified; the culture broth was centrifuged and the supernatant was extracted with ethyl acetate, and the organic layer was collected. The active fraction was evaporated and dissolved in methanol, and then silica gel col- umn chromatography was carried out; the active fraction was evaporated and dissolved in dichloromethane: methanol (1:5) mixture and Sephadex LH-20 column chromatography was carried out. The active fraction was evaporated and dissolved in water and lyophilized. The powdery antibiotic obtained in the above was dissolved in methanol and stored at 4 °C, and the crystalline antibiotic, trihexocin, was prepared; trihexocin had an antifungal activity against the plants infected with fungi at the concentration of 50-100 µg/ml. Tihexocin consisted of three fructose units and showed a non-toxicity.
La présente invention se rapporte à un nouveau micro-organisme Bacillus sp. SY-414 qui a été utilisé pour produire un antibiotique antifongique, la trihexocine, ainsi qu'à un procédé destiné à la fabrication de ce dernier. Le nouvel antibiotique antifongique répond à la formule générale (1), dans laquelle R représente -H, -OH, -NH2, un groupe alkyle C1-C6, un groupe allyle C5-C20 ou un groupe alkylène C2-C6, et l'indice l, m ou n représente un entier inférieur à 3. Le procédé de fabrication de l'antibiotique antifongique décrit ci-dessus est comme suit: un nouveau micro-organisme Bacillus sp. présentant une forte activité par rapport aux champignons infectant les végétaux, en particulier Pyricularia oryzae et Pellicularia filamentosa, a été isolé de la terre et identifié. Le bouillon de culture a été centrifugé et la substance surnageante extraite à l'aide de l'acétate d'éthyle, et la couche organique a été recueillie. La fraction active a été évaporée et dissoute dans du méthanol, après quoi la chromatographie sur colonne de gel de silice a été effectuée. La fraction active a été évaporée et dissoute dans un mélange dichlorométhane: méthanol (1:5) et la chromatographie sur Sephadex LH-20 a été effectuée. La fraction active a été évaporée et dissoute dans l'eau, puis lyophilisée. L'antibiotique en poudre obtenu selon les étapes susmentionnées a été dissous dans du méthanol et stocké à 4 DEG C, et l'antibiotique cristallin, la trihexocine, a ainsi été préparé.
