A CGTase having the following enzymological chemical properties: (1) action and substrate specificity: acting on starch, dextrin, amylopectin or amylose and thus forming mainly .gamma.-CD and relatively small amounts, compared with .gamma.-CD, of .beta.-CD and .alpha.-CD, (2) optimum pH: 10.5 to 11.0, (3) optimum temperature: around 60~C, (4) stable pH: 6 to 11, and (5) heat stability: showing a residual activity of 90% or more after treating at 50~C for 15 minutes; a process for producing CGTase which comprises culturing a microorganism belonging to the species Bacillus clarkii (for example, FERM BP- 7156) and collecting the CGTase thus produced in the culture medium; a process for producing CD which comprises treating a solution containing starch, etc. with CGTase produced by Bacillus clarkii to thereby produce mainly .gamma.-CD; a .gamma.-cyclodextrin glucanotransferase (CGTase) capable of preferentially producing .gamma.-CD; a microorganism capable of producing .gamma.-CGTase; and a process for producing .gamma.-CD by using .gamma.-CGTase.

L'invention concerne une CGTase possédant les propriétés chimiques enzymatiques suivantes : (1) action et spécificité du substrat : action sur l'amidon, la dextrine, l'amylopectine ou l'amylose et donc formation de cyclodextrine-.gamma., principalement, et de quantités relativement faibles, par rapport à la cyclodextrine-.gamma., de cyclodextrine-.beta. et de cyclodextrine-.alpha., (2) pH optimum : entre 10,5 et 11, (3) température optimum : environ 60 ·C, (4) pH stable : entre 6 et 11, et (5) stabilité à la chaleur : présente une activité résiduelle d'au moins 90 % après traitement à 50 ·C pendant 15 minutes. L'invention concerne également un procédé de production de CGTase consistant à cultiver un micro-organisme appartenant à l'espèce <i>Bacillus clarkii</i> (par exemple, FERM BP-7156) et à recueillir la CGTase ainsi produite dans le milieu de culture. L'invention concerne enfin un procédé de production de cyclodextrine consistant à traiter une solution contenant de l'amidon, etc. avec la CGTase produite par le <i>Bacillus clarkii</i> en vue de produire principalement de la cyclodextrine-.gamma., une CGTase de cyclodextrine-.gamma. capable de produire de manière préférentielle de la cyclodextrine-.gamma., un micro-organisme capable de produire de la CGTase-.gamma., ainsi qu'un procédé de production de cyclodextrine-.gamma. au moyen de CGTase-.gamma..
