The present invention relates to new optically pure compounds displaying an improved anticancer activity compared to previously known complex mixtures of stereoisomers thereof. Such compounds are of formula (I) : wherein each X independently represents any amino acid; n is 0 or 1; m is an integer between 0 and 3; k is an integer of at least 3; Psi is a reduced bond of formula replacing the peptide amide bond between Lys and Pro; and wherein Lys residues in pseudopeptide units of said compound of formula (I) are either all in L configuration or all in D configuration. A method for preparing such compounds, and therapeutic uses thereof are also provided. The synthetic method involves the selective reduction of the peptide bond between Lys and Pro in a dipeptide intermediate with borane. The therapeutic uses are against cancer, inflammation and for wound healing.
La présente invention porte sur de nouveaux composés optiquement purs qui présentent une activité anticancéreuse améliorée par comparaison avec des mélanges complexes précédemment connus de stéréo-isomères de ceux-ci. De tels composés sont de la formule (I) : dans laquelle les X représentent chacun indépendamment tout acide aminé ; n vaut 0 ou 1 ; m est un entier entre 0 et 3 ; k est un entier d'au moins 3 ; Psi est une liaison réduite de formule remplaçant la liaison peptide amide entre Lys et Pro, et les résidus Lys dans des unités de pseudopeptide dudit composé de la formule (I) étant soit tous en configuration L, soit tous en configuration D. L'invention porte également sur un procédé de préparation de tels composés, et sur les utilisations thérapeutiques de ceux-ci. Le procédé synthétique entraîne la réduction sélective par le borane de la liaison peptidique entre Lys et Pro dans un intermédiaire dipeptide. Les utilisations thérapeutiques sont contre le cancer, les inflammations et pour la cicatrisation de plaies.
