The invention relates to a method of synthesizing ivabradine of formula (I): (see formula I), the salts thereof for addition to a pharmaceutically acceptable acid and the hydrates thereof characterized in that a compound of formula (V) (see formula V) wherein R1 and R2, identical or different, each represents an alkoxy group (C1-C8) alkyl, or form together with the carbon atom that bears a 1,3 -dioxane, 1,3-dioxolane or 1,3-dioxepane to catalytic hydrogenation to yield the compound (VI) (see formula VI) is treated with a compound (VII) (see formula VII) in the presence of hydrogen and a catalyst to give the addition salt of HX to ivabradine or ivabradine if treated with base. The invention also provides a crystalline form of ivabradine hydrochloride.
L'invention concerne un procédé de synthèse de l'ivabradine de formule (I): (voir formule I) de ses sels d'addition à un acide pharmaceutiquement acceptable et de ses hydrates caractérisé en ce que l'on soumet un composé de formule (V) (voir formule V) dans laquelle R1 et R2, identiques ou différents, représentent chacun un groupement alkoxy (C1- C8) linéaire ou ramifié, ou bien forment ensemble avec l'atome de carbone qui les porte un cycle 1,3-dioxane, 1,3-dioxolane ou 1,3-dioxépane à une hydrogénation catalytique pour conduire au composé (VI) (voir formule VI) qui est traité avec un composé (VII) (voir formule VII) en présence d'hydrogène et un catalyseur pour conduire au sel d'addition de HX à l'ivabradine ou à l'ivabradine libre s'il est traité avec une base. L'invention concerne également la forme cristalline a du chlorhydrate de l'ivabradine.
