The invention relates to sigma receptor-deficient, mutant, non-human mammals and the applications thereof. The genome of the endogenous sigma receptor- deficient, mutant, non-human mammal contains a mutation comprising a disruption in a gene of an endogenous sigma receptor, whereby said gene disruption produces a mutant lacking detectable levels of endogenous sigma receptor. The aforementioned mutant can be used as a control animal for in vivo studies and as a source of cells to be used in in vitro studies. The sigma-1 receptor-deficient mutants can be used as models for the in vivo study of central nervous system disorders, memory alterations, stress conditions and drug addiction, neuroprotection and analgesic methods. The sigma-2 receptor- deficient mutants can be used to study diagnostic and therapeutic tools, in order to combat cancer and/or degenerative processes and/or to develop compounds that can prevent, reduce or alleviate the secondary pathology associated with the administration of neuroleptic agents.
Le génome du mammifère non humain mutant, à récepteurs sigma endogènes déficients contient une mutation comprenant le clivage en un gène d'un récepteur sigma endogène, ledit clivage génique donnant lieu à un mutant exempt de taux détectables du récepteur sigma endogène. Le mutant peut être utilisé comme témoin dans des essais biologiques <i>in vivo</i> ainsi que comme source cellulaire susceptible d'être utilisée dans des essais biologiques <i>in vitro</i>. Les mutants à récepteur sigma-1 déficient peuvent servir de modèles pour l'étude <i>in vivo</i> de troubles du système nerveux central, d'altérations de la mémoire, d'états de stress et de dépendance à des drogues, de processus d'analgésie et de neuroprotection. Les mutants à récepteur sigma-2 déficient peuvent servir à étudier les ustensiles, les diagnostics ou thérapies permettant de lutter contre le cancer et/ou des processus dégénératifs et/ou à concevoir des composés aptes à prévenir, réduire ou soulager la pathologie secondaire associée à l'administration d'agents neuroleptiques.
