The invention relates to novel benzodiazepine derivatives with antiproliferative activity and more specifically to novel benzodiazepines of formula (I) and (II), in which the diazepine ring (B) is fused with a heterocyclic ring (CD), wherein the heterocyclic ring is bicyclic or a compound of formula (III), in which the diazepine ring (B) is fused with a heterocyclic ring (C), wherein the heterocyclic ring is monocyclic. The invention provides cytotoxic dimers of these compounds. The invention also provides conjugates of the monomers and the dimers. The invention further provides compositions and methods useful for inhibiting abnormal cell growth or treating a proliferative disorder in a mammal using the compounds or conjugates of the invention. The invention further relates to methods of using the compounds or conjugates for in vitro, in situ, and in vivo diagnosis or treatment of mammalian cells, or associated pathological conditions.
La présente invention porte sur de nouveaux dérivés de benzodiazépine présentant une activité antiproliférative, et concerne plus particulièrement les nouveaux dérivés de benzodiazépine des formules (I) et (II), le cycle de diazépine (B) étant dans ce cas fusionné avec un cycle hétérocyclique (CD) et le cycle hétérocyclique étant bicyclique, ou un composé de la formule (III), le cycle de diazépine (B) étant dans ce cas fusionné à un cycle hétérocyclique (C) et le cycle hétérocyclique étant monocyclique. L'invention porte sur des dimères cytotoxiques desdits composés. L'invention se rapporte également à des conjugués de monomères et de dimères. L'invention concerne en outre des compositions et procédés utiles pour inhiber une croissance cellulaire anormale ou pour traiter un trouble prolifératif chez un mammifère à l'aide des composés ou conjugués de l'invention. L'invention porte enfin sur des procédés d'utilisation desdits composés ou conjugués pour le diagnostic ou le traitement in vitro, in situ et in vivo de cellules de mammifères ou d'autres états pathologiques associés.
