The role for a substantially purified human growth factor preparation is shown to contain a distinct polypeptide with apparent Mr on SDS/PAGE of about 220 kD, and to be distinct from bFGF, with a most active fraction at about 220 kD. A human growth factor polypeptide of 157 kD was identified, in human bone marrow aspirates and from bone stromal cells, by immunoblotting with the mAb MS 329, and by affinity isolation using an MS 329 antibody column. A PSA stimulating autocrine factor, PSAF, that is precipitable in ammonium sulfate at between 60-80 % saturation is able to induce PSA expression and secretion, and serves as an indicator of androgen independent prostate cell growth.

Une préparation très purifiée à base de facteur de croissance humain se révèle contenir un polypeptide distinct d'un poids moléculaire apparent d'environ 220 kD détecté par SDS-page et distinct du bFGF, avec une fraction très active d'environ 220 kD. Un polypeptide du facteur de croissance humain de 157 kD a été identifié dans des produits d'aspiration de moelle osseuse humaine et dans des cellules stromales osseuses par immunotransfert à l'aide du mAb MS 329 et par isolement par affinité à l'aide d'une colonne d'anticorps MS 329. Un facteur autocrine stimulant l'antigène spécifique de la prostate (PSAF) précipitable dans le sulfate d'ammonium, pour une saturation comprise entre 60 et 80 %, est capable d'induire l'expression et la sécrétion de l'antigène spécifique de la prostate et sert d'indicateur de croissance des cellules prostatiques indépendantes des androgènes.
