Brevibacillus choshinensis exhibits strikingly low proteolytic activity to cell outside and also exhibits excellent performance in the capability of protein secretion and production. However, it is desired to not only further reduce the extracellular proteolytic activity thereof but also achieve a reduction of intracellular proteolytic activity. Moreover, in the use of this bacterium as a host for protein drugs, etc., it is desired that the bacterium be easily destroyed without spore formation. These have been resolved by inactivating spore formation associated genes and by with respect to extracellular and intracellular protease genes, carrying out cloning and inactivation.
L'invention concerne la bactérie Brevibacillus choshinensis qui présente une activité protéolytique extracellulaire extrêmement faible ainsi qu'une excellente capacité de sécrétion et de production de protéines. L'invention n'a pas pour seul objectif de réduire l'activité protéolytique extracellulaire de cette bactérie mais également son activité protéolytique intracellulaire. De plus, lorsque cette bactérie est utilisée en tant qu'hôte pour des médicaments protéiniques, etc., l'invention a également pour objectif de détruire facilement cette bactérie sans entraîner la formation de spores. Pour résoudre ces problèmes, les gènes associés à la formation de spores ont été inactivés par rapport aux gènes de la protéase extracellulaires et intracellulaires, ce qui permet de procéder au clonage et à l'inactivation de ces derniers.
