The present invention relates to novel vectors, to DNA vaccines and gene therapeutics containing said vectors, to methods for the preparation of the vectors and DNA vaccines and gene therapeutics containing the vectors, and to therapeutic uses of said vectors. More specifically, the present invention relates to novel vectors comprising (a) an expression cassette of a gene of a nuclear-anchoring protein, which contains (i) a DNA binding domain capable of binding to a specific DNA sequence and (ii) a functional domain capable of binding to a nuclear component and (b) a multimerized DNA sequence forming a binding site for the anchoring protein, and optionally (c) one or more expression cassettes of a DNA sequence of interest. In particular the invention relates to vectors that lack a papilloma virus origin of replication. The nuclear-anchoring protein might be the E2 protein of Bovine Papilloma Virus type 1 or Epstein-Barr Virus Nuclear Antigen 1. The invention also relates to vectors that lack an origin of replication functional in a mammalian cell. The invention further relates to methods for expressing a DNA sequence of interest in a subject.
La présente invention concerne de nouveaux vecteurs, des vaccins d'ADN et des agents thérapeutiques géniques renfermant lesdits vecteurs, des méthodes de préparation desdits vecteurs, vaccins d'ADN et agents thérapeutiques, ainsi que des utilisations thérapeutiques de ces vecteurs. Plus spécifiquement, cette invention a trait à de nouveaux vecteurs renfermant (a) une cassette d'expression d'un gène d'une protéine d'ancrage nucléaire qui contient (i) un domaine de liaison à l'ADN capable de se lier à une séquence d'ADN spécifique et (ii) un domaine fonctionnel capable de se lier à un composé nucléaire, et (b) une séquence d'ADN multimèrisé formant un site de liaison destiné à la protéine d'ancrage, et facultativement (c) au moins une cassette d'expression d'une séquence d'ADN d'intérêt. Notamment, cette invention a pour objet des vecteurs exempts d'une origine de réplication du papillomavirus. Cette protéine d'ancrage nucléaire peut être la protéine E2 de type du papillomavirus bovin ou un antigène 1 nucléaire du virus d'Epstein-Barr. La présente invention concerne, en outre, des vecteurs exempts d'une origine de réplication fonctionnelle dans une cellule mammalienne, ainsi que des méthodes d'expression d'une séquence d'ADN d'intérêt chez un sujet.
