Novel farnesyl transferase inhibitors of general formula (I), preparation thereof and pharmaceutical compositions containing same. In general formula (I), R1 is Y-S-A1 (where Y is a hydrogen atom, an amino acid residue, a fatty acid residue, or an alkyl or alkoxycarbonyl radical, and A1 is a C1-4 alkylene radical optionally alpha -substituted in the >C(X1)(Y1) grouping by an amino, alkylamino, alkanoylamino or alkoxycarbonylamino radical wherein the alkyl or alkanoyl portion contains 1-6 carbon atoms; each of X1 and Y1 is a hydrogen atom or X1 and Y1, taken together with the carbon atom to which they are attached, form a >C=O grouping; R'1 is hydrogen or a C1-6 alkyl radical; X is an oxygen or sulphur atom; R2 is a C1-6 alkyl, alkenyl or alkynyl radical optionally substituted by hydroxy, alkoxy, mercapto, alkylthio, alkylsulphinyl or alkylsulphonyl, with the proviso that when R2 is an alkyl radical substituted by a hydroxy radical, then R2 and the alpha carboxy radical can form a lactone; R'2 is hydrogen or a C1-6 alkyl radical; and R is a hydrogen atom, an optionally substituted alkyl radical or an optionally substituted phenyl radical. Said novel products have anticancer properties.

Nouveaux inhibiteurs de farnésyl transférase de formule générale (I), leur préparation et les compositions pharmaceutiques qui les contiennent. Dans la formule générale (I), R1 représente Y-S-A1-(Y = atome d'hydrogène, reste d'aminoacide, reste d'acide gras, radical alkyle ou alcoxycarbonyle, et A1 = radical alcoylène contenant 1 à 4 atomes de carbone éventuellement substitué en a du groupement >=C(X1)(Y1) par un radical amino, alkylamino, alkanoylamino, alkoxycarbonylamino dont la partie alkyle ou alkanoyle contient 1 à 6 atomes de carbone, X1 et Y1 représentent chacun un atome d'hydrogène ou forment ensemble avec l'atome de carbone auquel ils sont liés un groupement > C=O, R'1 représente hydrogène ou un radical alkyle contenant 1 à 6 atomes de carbone, X représente un atome d'oxygène ou de soufre, R2 représente un radical alkyle, alcényle ou alcynyle contenant 1 à 6 atomes de carbone éventuellement substitué par hydroxy, alkoxy, mercapto, alkylthio, alkysulfinyle ou alkylsufonyle, étant entendu que, lorsque R2 représente un radical alkyle substitué par un radical hydroxy, R2 peut former avec le radical carboxy en .alpha. une lactone, R'2 représente hydrogène ou un radical alkyle contenant 1 à 6 atomes de carbone, et R représente un atome d'hydrogène ou un radical alcoyle éventuellement substitué ou un radical phényle éventuellement substitué. Ces nouveaux produits présentent des propriétés anticancéreuses.
