Novel structural forms of T cell costimulatory molecules are described. These structural forms comprise a novel structural domain or have a structural domain deleted or added. The structural forms correspond to naturally-occuring alternatively spliced forms of T cell costimulatory molecules or variants thereof which can be produced by standard recombinant DNA techniques. In one embodiment, the T cell costimulatory molecule of the invention contains a novel cytoplasmic domain. In another embodiment, the T cell costimulatory molecule of the invention contains a novel signal peptide domain or has an immunoglobulin variable region-like domain deleted. The novel structural forms of T cell costimulatory molecules can be used to identify agents which stimulate the expression of alternative forms of costimulatory molecules and to identify components of the signal transduction pathway which results in costimulation of T cells.
L'invention concerne de nouvelles formes structurelles de molécules costimulantes des lymphocytes T. Ces formes structurelles comprennent un nouveau domaine structurel ou possèdent un domaine structurel supprimé ou ajouté, et elles correspondent à des formes d'origine naturelle, à épissage alternatif, de molécules costimulantes de lymphocytes T ou de variantes de celles-ci que l'on peut produire à l'aide de techniques courantes d'ADN recombiné. Dans un mode de réalisation de l'invention, la molécule costimulante de lymphocytes T contient un nouveau domaine cytoplasmique. Dans un autre mode de réalisation, cette molécule contient un nouveau domaine de peptide-signal, ou on a supprimé dans celle-ci un domaine de type à région variable d'immunoglobuline. On peut utiliser ces nouvelles formes structurelles de molécules costimulantes de lymphocytes T pour identifier des agents stimulant l'expression de formes alternatives de molécules costimulantes et pour identifier des constituants du chemin de transduction du signal ayant pour effet de costimuler les lymphocytes T.
