The present invention deals with a modified polypeptide comprising three contiguous segments N, L and C represented by the formula N-L-C and comprising : a N-helix region of gp41 (N), a C-helix region of gp41 (C), and a connecting loop comprising a synthetic linker (L) between the N and C-helices, the linker replacing amino acids 593 - 617 of gp41, the numbering scheme being based upon the prototypic isolate HIV-1 HxB2 Clade B strain, said polypeptide comprising the calveolin-1 neutralizing and 98.6 D epitopes, but not 2F5 and 4E10 epitopes, not the fusion peptide, the polypeptide having a minimal immunogenic cross-reactivity with human interleukin 2(IL2).
La présente invention concerne un polypeptide modifié comprenant trois segments contigus, N, L et C, représentés par la formule N-L-C et comprenant: une région d'hélice N de gp41 (N); une région d'hélice C de gp41 (C); et une boucle de liaison comprenant un lieur synthétique (L) entre les hélices N et C, le lieur remplaçant les acides aminés 593 à 617 de gp41, le plan de numérotage étant basé sur l'isolat prototypique de la souche HxB2 du clade B du VIH-1, ledit polypeptide comprenant les épitopes neutralisants la calvéoline-1 et les épitopes 98.6 D, mais pas les épitopes 2F5 et 4E10, ni le peptide de fusion, ledit polypeptide présentant une réactivité croisée immunogène minimale avec l'interleucine 2 (IL2) humaine.
