A full-length cDNA encoding a human-origin G protein-coupled receptor protein is isolated by screening a human hippocampus library. Also, a rat-origin cDNA corresponding to the above human-origin cDNA is isolated. Proteins encoded by these cDNAs have an activity of lowering intracellular cAMP concentration under stimulation with histamine. These proteins are usable as tools in screening ligands thereof or in screening candidate compounds for drugs capable of regulating signal transduction from the above proteins.
On isole, par criblage d'une bibiliothèque de l'hippocampe humain, un ADN complémentaire complet codant pour une protéine réceptrice d'origine humaine couplée à la protéine G. On isole également un ADN complémentaire issu d'un rat correspondant à l'ADN complémentaire humain mentionné ci-dessus. Les protéines codées par ces ADN complémentaires diminuent la concentration d'AMP cyclique intracellulaire par stimulation à l'histamine. Ces protéines peuvent être utilisées comme outils pour le criblage de leurs ligands ou pour le criblage de composés candidats pour obtenir des médicaments capables de réguler la transduction de signal desdites protéines.
