Described herein is a variant human acidic mammalian chitinase (AMCase) enzyme having improved stability and an isolated nucleic acid sequence which encodes the variant. The invention also provides to nucleic acid constructs, vectors, and host cells comprising the nucleic acid sequences as well as methods for producing recombinant variant AMCase polypeptide and its use in screening assays to identify modulators.

L'invention porte sur une enzyme chitinase mammalienne acide humaine variante ("acidic mammalian chitinase" ou AMCase) à stabilité améliorée et sur une séquence nucléotidique qui code le variant. L'invention se rapporte également à des constructions d'acides nucléiques, à des vecteurs et des cellules hôtes renfermant les séquences nucléotidiques précitées, à des procédés qui permettent de produire le polypeptide AMCase variant recombinant et à leur utilisation dans des épreuves de criblage permettant d'identifier des modulateurs.
