A cancer/testis antigen applicable to the diagnosis and immunotherapy for cancer, namely, a cancer-testis antigen which is not expressed in normal tissues except testis but expressed in various cancers over a broad range and thus induces an immune reaction in a host; a cancer/testis gene encoding the same, etc. By using a yeast two-hybrid system, a protein binding specifically to GCF, which has been reported as a transcriptional regulatory factor, is screened. Then a gene of this protein specifically binding to GCF is cloned. It is confirmed that protein D40, which is the gene product thereof, is a cancer/testis antigen not substantially expressed in human normal tissues except testis but is expressed in human primary cancers originating in various tissues and cells. Further, it is found out that D40 has a high affinity for HLAs and has a sequence consisting of 9 or 10 amino acid residues binding to a plural number of HLAs.

L'invention concerne un antigène de type cancer-testis pouvant être utilisé dans le diagnostic et l'immunothérapie du cancer, plus spécifiquement, un antigène de type cancer-testis qui n'est pas exprimé dans les tissus normaux excepté les testicules, mais qui est exprimé dans une grande variété de cancers différents et qui induit une réaction chez un hôte ; ainsi qu'un gène de type cancer-testis codant pour cet antigène, etc. Au moyen d'un système double-hybride de levure, on crible une protéine se liant spécifiquement au facteur de liaison d'élément GC (GCF), lequel a été présenté comme un facteur de régulation transcriptionnel. Ensuite, un gène de cette protéine se liant spécifiquement au GCF est cloné. On confirme que la protéine D40, qui est le produit génétique de celui-ci, est un antigène de type cancer-testis qui n'est pas sensiblement exprimé dans les tissus normaux humains, excepté dans les testicules, mais qui est exprimé dans des cancers primaires humains ayant leur origine dans différents tissus et cellules. En outre, on a découvert que D40 possède une grande affinité pour les antigènes HLA et possède une séquence consistant en 9 ou 10 résidus d'aminoacides se liant à plusieurs antigènes HLA.
