The present invention provides an efficient and novel maize transformation and regeneration system based on a novel split-seed explant. Mature maize seeds are split longitudinally to form a split-seed explant. The split-seed explant can then be used in transformations to introduce a gene of interest into the maize genome to produce novel maize lines having desired characteristics. The split-seed explant can also be used to generate calli and/or multiple shoots, and rooted plantlets.

L'invention porte sur une nouveau et efficace système de transformation et régénération du maïs basé sur un nouvel explant de graine fendue. A cet effet on fend longitudinalement des graines de maïs mûres pour former des explants de graines fendues qui peuvent servir lors de transformations pour introduire un gène d'intérêt dans le génome du maïs afin de créer de nouvelles lignées de maïs présentant les caractéristiques désirées. On peut également utiliser les explant de graines fendues pour créer des calli et/ou des pousses multiples ou des plantules radiculées.
