The invention relates to a method for generating a DNA sequence coding for the heavy chain or the light chain of at least one antibody from RNA from a cell capable of producing an antibody. More particularly, the invention relates to the generation of a monoclonal antibody library. The invention also relates to the use of an antibody library for screening monoclonal antibodies, preferably human antibodies for treating cancer.
La présente invention vise un procédé de génération d'une séquence d'ADN codant pour une chaîne lourde ou une chaîne légère d'au moins un anticorps à partir d'ARN issu d'une cellule capable de produire un anticorps. Plus particulièrement, l'objet de l'invention vise la génération d'une banque d'anticorps monoclonaux. L'invention concerne également l'utilisation d'une banque d'anticorps pour le criblage d'anticorps monoclonaux, préférentiellement d'anticorps humains destinés au traitement du cancer.
