The present invention provides two embodiments. One is a method of testing for compounds which may induce STF-1 transcription. Pancreatic islet cells which express the STF-1/lacZ fusion gene are isolated and the effect of various compounds on STF-1 expression is measured by adding the compound of interest to STF-1/lacZ expressing cells. LacZ activity in control and treated cells is then quantitated by a colorimetric assay. Using this method, a large number of compounds can be screened and STF-1 inducing compounds can be identified readily. The other is a method of using the STF-1 promoter to mark insulin producing pancreatic islet cells in vivo. In this regard, the green fluorescent protein (GFP) served as an indicator. The introduction of the STF-1-green fluorescent protein transgene into pigs allows for the efficient and rapid recovery of insulin producing cells from the pancreas.

La présente invention concerne deux réalisations. La première réalisation consiste en un procédé d'analyse de composés susceptibles d'induire la transcription de la protéine STF-1. On isole des cellules d'îlots pancréatiques qui expriment le gène de fusion STF-1/lacZ, et on mesure l'effet des différents composés sur l'expression de STF-1 en ajoutant le composé étudié aux cellules exprimant STF-1/lacZ. Un dosage par procédé colorimétrique permet alors de quantifier l'activité de lacZ dans des cellules traitées et dans des cellules témoin. Ce procédé permet de vérifier un grand nombre de composés, et d'identifier rapidement les composés induisant la protéine STF-1. L'autre réalisation consiste en un procédé d'utilisation d'un promoteur de STF-1 pour marquer in vivo les cellules d'îlots pancréatiques producteurs d'insuline. Dans ce cadre, c'est la protéine fluorescente verte (GFP) qui a servi d'indicateur. L'introduction chez le porc du transgène de STF-1 et de la protéine fluorescente verte a permis une guérison efficace et rapide des cellules du pancréas produisant de l'insuline.
